CRISPR Cas9/gRNA RNP（核糖核蛋白复合物）由CRISPR RNA(crRNA)和tracrRNA部分双链RNA复合物及cas蛋白组成。不同于传统的Cas9/gRNA质粒，crRNA和tracrRNA寡聚核苷酸是完整的组分，不需要细胞自身转录得到CRISPR相关组分。直接将Cas9protein 跟sgRNA混合就可以用于转染或者注射。Cas9 RNP与其它系统相比有多个优点：
1）Cas9 RNP在转染后即刻发生高效率基因编辑
2）Cas9 RNP通过蛋白降解可快速清除以降低脱靶可能性
3）Cas9 RNP是in vivo基因编辑的完美方案
