纳米粒子广泛用于医学诊断,特别是黄金纳米粒子(AuNPs),由于其特殊的光学特性,更是广泛地作为生物传感器运。当AuNPs 溶液中存在PNA时,由于PNA是中性电荷的,PNA 起到了凝集剂的作用,促使AuNPs 聚合。 当AuNPs聚合时,AuNPs粒子会发生光学变化,如颜色改变(红色变成暗紫色)。利用这个原理,可以检测样品中是否存在某一特定核酸序列,从而检测是否有相应的病原体。
如果溶液中存在跟PNA互补的RNA片段,则PNA会跟RNA配对。这时PNA就不能促使AuNPs 凝集,颜色不会变成暗紫色。
1 PNA单独存在时,能够促使AuNP 凝集(暗紫色)。
2 当存在与PNA互补的RNA时,PNA与RNA结合,AuNP处于分散状态(红色)
3 当溶液中的RNA与PNA不互补时,PNA不会跟RNA特异性结合,AuNP还是会凝集(暗紫色)
